SOP FOR OPERATION & CALIBRATION OF UV CABINET

  1. OBJECTIVE:

    1. To lay down the procedure for Operation & Calibration of UV Cabinet.

  2. SCOPE

    1. This procedure is applicable for the Operation & Calibration of UV Cabinet of QC Laboratory 

  3. RESPONSIBILITY:

    1. Quality Control Executive is responsible to follow procedure as per the procedure mentioned in this SOP. 

    2. QC Head is responsible for implementation and compliance of the procedure as per this SOP.

  4. ABBREVIATIONS

    3. UV    :    Ultraviolet

    4. TLC    :    Thin layer chromatography

  5. PROCEDURE

    1. Operating Procedure.

  1. Ensure that the instrument is clean and free from dust.

  2. Switch ‘ON’ the mains.

  3. Switch ‘ON’ the instrument.

  4. Open the cabinet and place the TLC plate inside the cabinet and close the door.

  5. Three switches are provided on the top corner of the instrument.

  • Switch 1- For visible light

  • Switch 2 – For UV light at short wavelength (254)

  • Switch 3- For UV light at long wave length (365)

    1. Select the required UV light and switch it ON.

    2. Observe the plate for visualization of spot from the observation compartment (window).

    3. Switch ‘OFF’ the instrument when not in use.

    1. PRECAUTIONS

  1. Proper handling of the instrument.

  2. Do not stare at the UV rays for a long time.

  3. Do not use organic solvent for cleaning the instrument

    1. Calibration

  1. Calibration of UV cabinet is done by using 0.2% w/v solution & 0.04% w/v solution of sodium salicylate in 95% ethanol, observed at 254 & 365 nm respectively.

  2. Prepare solution of 0.2% and 0.04% solution if sodium salicylate in 95% ethanol.

  3. Apply a spot of the above solution on two different TLC plates and let the spots dry.

  4. Select the required UV light and switch it ON.

  5. Place the TLC plate & observe the plate for visualization of spot from the observation compartment (window).

    1. Frequency of Calibration

    1. Quarterly.
