SOP FOR OPERATION AND CALIBRATION OF POLARIMETER

  1. OBJECTIVE:

    1. To lay down the procedure for Operation and Calibration of Polarimeter.

  2. SCOPE

    1. This procedure is applicable for the Operation and Calibration of Polarimeter in the lab.

  3. RESPONSIBILITY:

    1. Quality Control Officer / Executive are responsible to follow procedure as per the procedure mentioned in this SOP. 

    2. QC Head is responsible for implementation and compliance of the procedure as per this SOP.

  5. PROCEDURE

    1. Procedure For Operation

      1. Ensure that the working bench and instrument is clean.

      2. Switch on the instrument and allow to stabilize for 30 minutes.

      3. Fill the Polarimeter tube with a clear solution of the sample prepared as mentioned in respective testing procedure; in such a way as to avoid air bubbles.

      4. Tighten the end plates to ensure a leak proof seal.

      5. Observe the optical rotation angle directly from the scale.

      6. Determine the specific optical rotation by the formula 

SOR = α 100 / CL

Where α is degree of rotation at specified temperature.

C is the concentration in gram per ml.

L is the length of tube in decimeters.

    1. Calibration Procedure

      1. Operate the instrument as mentioned in procedure for operation.

      2. Prepare the solutions of sucrose AR grade as mentioned in table and observe the optical rotation.

      1. Calibration Frequency – Quarterly. Put a calibration tag after calibration.

      2. If the instrument is out of calibration tag the instrument with UNDER MAINTENANCE   label.

      3. Inform to the service engineer for rectification. Calibrate the instrument after rectification.
